Lactoferrin, an iron-binding protein found in high concentrations in mammalian exocrine secretions, is an important component of the host defense system. It is also a major protein of the secondary granules of neutrophils from which is released upon activation. Due to its potential clinical utility, recombinant human lactoferrin (rhLF) has been produced in various eukaryotic expression systems; however, none of these are fully compatible with humans. Most of the biopharmaceuticals approved by the FDA for use in humans are produced in mammalian expression systems. The Chinese hamster ovary cells (CHO) have become the system of choice for proteins that require post-translational modifications, such as glycoproteins.  相似文献

Microorganisms that survive in natural environments form organized multicellular communities, biofilms and colonies with specific properties. During stress and nutrient limitation, slow growing and senescent cells in such communities retain vital processes by maintaining plasma membrane integrity and retaining the ability to generate transmembrane electrochemical gradients. We report the use of a Saccharomyces cerevisiae colonial model to show that population growth in a multicellular community depends on nutrient diffusion and that resting cells start to accumulate from the beginning of the second acidic phase of colony development. Despite differentiation of colony members, synchronous transmembrane potential oscillation was detected in the organized colony. The electrochemical membrane potential periodically oscillated at frequencies between those for circadian to infradian rhythms during colony aging and transiently decreased at time points previously linked with rebuilding of yeast metabolism. Despite extensive decreases in the intracellular ATP concentration and in the amount and activity of the plasma membrane proton pump during nutrient limited growth and colony aging, the transmembrane electrochemical potential appeared to be maintained above a level critical for population survival.  相似文献

采用光学显微镜对旱生金毛裸蕨(Gymnopteris vestita)配子体发育的全过程进行了观察。结果显示,(1)旱生金毛裸蕨孢子三裂缝,成熟时黄褐色,接种后10~15d萌发,萌发类型为书带蕨型。原叶体母细胞首先形成单列的丝状体,其后配子体发育明显区别于非旱生的蕨类,金毛裸蕨配子体发育最明显的特征是形成大量的分枝,通常单列的丝状体基部细胞可通过细胞纵分裂形成丝状分枝,这些分枝又可进一步产生新的分枝,分枝的末端可形成片状体,这些片状体又可产生分枝丝状体或片状体,最终整个配子体可发育为群丛。有时,单列的丝状体也可直接发育为片状体,然而这些片状体并不发育为原叶体而是产生大量的丝状分枝。当群丛形成时,在丝状体或片状体表面可产生数量较多的精子器,但在人工培养条件下并没有发现颈卵器。如果培养条件适宜,配子体可进行营养繁殖,持续较长时间,老的片状体上可产生新的丝状体。金毛裸蕨位于群丛外的大型心形原叶体可进行无配子生殖产生孢子体。金毛裸蕨的配子体发育特征,包括多分枝、发达的营养繁殖及无配子生殖现象的发生,表明了金毛裸蕨配子体群丛的形成是对于旱生环境的一种适应性。  相似文献

Maximum Likelihood (ML) method has an excellent performance for Direction-Of-Arrival (DOA) estimation, but a multidimensional nonlinear solution search is required which complicates the computation and prevents the method from practical use. To reduce the high computational burden of ML method and make it more suitable to engineering applications, we apply the Artificial Bee Colony (ABC) algorithm to maximize the likelihood function for DOA estimation. As a recently proposed bio-inspired computing algorithm, ABC algorithm is originally used to optimize multivariable functions by imitating the behavior of bee colony finding excellent nectar sources in the nature environment. It offers an excellent alternative to the conventional methods in ML-DOA estimation. The performance of ABC-based ML and other popular meta-heuristic-based ML methods for DOA estimation are compared for various scenarios of convergence, Signal-to-Noise Ratio (SNR), and number of iterations. The computation loads of ABC-based ML and the conventional ML methods for DOA estimation are also investigated. Simulation results demonstrate that the proposed ABC based method is more efficient in computation and statistical performance than other ML-based DOA estimation methods.  相似文献

目的:分析并总结食管破裂的患者急诊住院后经过系统、科学、快速的护理手段和临床治疗,及使用特殊的治疗方法(借助电子胃镜行食管破裂的修补治疗)得到救治的过程中的护理体会.方法:入院后急检血常规、血型、凝血象、生化系列等相关检查;给予禁食水,重症监护,对症处理,营养支持,胸腔闭式引流术排出引流液,应用敏感抗生素等治疗方法;给予科学的严密护理(生命体征的监测护理、严密观察体温的变化及对症处理、胃管的护理、胸腔闭式引流管的护理、心里支持的护理).结果:通过上述方法,患者在无明显并发症发生的情况下,最终在较短的时间内顺利痊愈出院.结论:与常规开胸下行食管破裂修补术治疗食管破裂的患者相比,在电子胃镜下行食管破裂修补术拥有治疗时间短,恢复快,住院时间短,患者医疗费降低等优点.  相似文献

The Tinhosas islands, in São Tomé e Príncipe, host the most important seabird breeding colony in the Gulf of Guinea, but information on its conservation status was hitherto unpublished or anecdotal, the last assessment having been performed in 1997. A two-day expedition to the Tinhosas islands was undertaken to estimate the status of breeding seabirds in 2013. Four of the five seabird species known to breed in São Tomé e Príncipe, namely Brown Booby Sula leucogaster, Sooty Tern Onychoprion fuscatus, Brown Noddy Anous stolidus and Black Noddy Anous minutus, occur on the Tinhosas. A decrease of 80% in Brown Booby numbers, possibly due to occasional exploitation, and a 30% increase in Sooty Tern and Black Noddy numbers, were found compared to 1997 data although survey methods differed. Breeding of Brown Noddy and Madeiran Storm-petrel Hydrobates castro remains unconfirmed. Our estimates confirmed that BirdLife International Important Bird and Biodiversity Area criteria are met for at least one species, the Sooty Tern. The islands are not legally protected, nonetheless, apart from moderate levels of disturbance by fishermen who land on Tinhosa Grande, no alien species were seen, and no immediate threats to the Tinhosas colony were detected. Multiple visits within and between years are recommended, to census breeders, monitor threats and establish breeding phenologies.  相似文献

Bumble bee (Bombus) species are ecologically and economically important pollinators, and many species are in decline. In this article, we develop a mechanistic model to analyse growth trajectories of Bombus vosnesenskii colonies in relation to floral resources and land use. Queen production increased with floral resources and was higher in semi‐natural areas than on conventional farms. However, the most important parameter for queen production was the colony growth rate per flower, as opposed to the average number of available flowers. This result indicates the importance of understanding mechanisms of colony growth, in order to predict queen production and enhance bumble bee population viability. Our work highlights the importance of interpreting bumble bee conservation efforts in the context of overall population dynamics and provides a framework for doing so.  相似文献

目的:探讨闭合复位与切开复位对桡骨远端骨折患者的临床疗效。方法:回顾性分析2013年6月至2014年3月我院收治的60例桡骨远端骨折患者,随机数字表法分为切开复位组和闭合复位组,每组30例。闭合复位组患者给予闭合复位小夹板或石膏固定治疗,切开复位组患者给予切开复位内固定术治疗。观察并比较两组患者治疗前后掌倾角、尺偏角以及桡骨长度、术中出血量和手术时间,骨折愈合时间以及患者临床疗效进行检测并比较。结果:与治疗前相比,治疗后患者的掌倾角、尺偏角、桡骨长度水平均升高(P0.05);与闭合复位组相比,切开复位组患者掌倾角、尺偏角、桡骨长度评分水平较高(P0.05),术中出血量、手术时间水平较高(P0.05),临床治疗的优良率较高(P0.05),两组患者的骨折愈合时间无显著差异(P0.05)。结论:与闭合复位相比,切开复位能够明显恢复桡骨远端骨折患者的掌倾角、尺偏角以及桡骨长度,但手术时间以及术中出血量较多,临床疗效较好,两组患者的骨折愈合时间无明显差异。  相似文献

IntroductionCell damage inescapably occurs during both the freezing and the thawing graft processes for autologous hematopoietic stem cell (HSC) transplantation. To estimate HSC injury, a quality control is performed including: (i) CD34+ quantification; (ii) percentage of CD34+ viability and (iii) evaluation of HSC functional ability to form colony forming unit–granulocyte macrophage (CFU-GM). Apoptosis involves complex pathways such as caspase enzymes. Here, we assess the extent of apoptosis that is caspase-dependent before and after cryoconservation of CD34+, using a Fluorescent Labeled Inhibitor of CAspases (FLICA).MethodsCaspase pathway activation status was evaluated in 46 patients (multiple myeloma [n = 24], lymphoma [n = 22]), by flow cytometry, using a 7-aminoactinomycin-D (7AAD)/FLICA staining test, in CD34+, CD3+, CD14+ and CD56+ cells. Viable 7AAD?/FLICA+ cells were then correlated with various parameters.ResultsWe showed a significant caspase pathway activation, with 23% CD34+/7AAD?/FLICA+ cells after thawing, compared with the 2% described in fresh CD34+ cells (P < 0.0001). Moreover, caspase pathway was significantly activated in thawing CD3+, CD56+ and CD14+ cells. We also report a significant correlation between the rate of CD34+/7AAD?/FLICA+ cells and post-thawing granulocytes count (P = 0.042) and their potential to be differentiated into CFU-GM (P = 0.004).DiscussionOur results show substantial cell death, induced by the increase of caspase pathway activation, secondary to the thawing process, and across all study cell types. This observation may affect the immune response quality during recipient aplasia, without detecting a clinical impact. Moreover, caspase pathway activation through CD3+ and CD56+ subpopulations could modify the therapeutic result of donor lymphocytes infusion (DLI).  相似文献
